During World War II, Mature's patriotic spirit led him to enlist in the U.S. Coast Guard after being rejected by the Navy due to color blindness. He served aboard the USCGC Storis and later on the troop transport USS Admiral H. T. Mayo, contributing to morale shows and War Bond tours to support the troops.

Post-war, Mature's acting career flourished as he took on diverse roles, showcasing his talent and range. He notably portrayed Doc Holliday in John Ford's "My Darling Clementine" alongside Henry Fonda, solidifying his place in Hollywood.

Mature's filmography includes iconic roles in Westerns, Biblical epics, and musicals. He starred in classics like "The Robe" and its sequel "Demetrius and the Gladiators," as well as "Samson and Delilah" and "The Egyptian," where he demonstrated his ability to embody complex characters with depth and charisma.
